Current Research Projects | 1) Multidrug resistance transporters are a large family of membrane proteins which play important roles in many physiological processes, disease as well as drug resistance. These proteins undergo large scale conformational changes as they translocate substances into or out of cells. Therefore the conformational dynamics of multidrug resistance transporters is crucial to their working mechanism. We use multi-scale molecular simulation techniques to explore the large-scale conformational change of transporters, aiming at unraveling their transport mechanisms at atomic level. 2) Asymmetric cell division (ACD) is a fundamental process to generate cellular diversity during animal development. In ACD, cells establish a polarity axis to coordinate the polarized distribution of cell fate determinants and orientation of mitotic spindle, giving rise to two daughter cells with different cell fates. We use structural biology and biochemical methods to characterize the protein complexes that function to regulate the spindle orientation and cell fate determinants segregation to understand the detailed molecular mechanism underlying ACD. |
